In the 1960s, superpowered humans Charles Xavier and Erik Lensherr work together to find others like them, but Erik's vengeful pursuit of an ambitious mutant who ruined his life causes a schism to divide them.

Film dibuka dengan sekuen pembuka film X-Men (2000) yang memperlihatkan Erik Lensherr kecil yang harus terpisah dari ibunya pada masa holocaust. Kisah berlanjut, ternyata kekuatan si bocah diamati Dr. Klaus Schmidt seorang ilmuwan Nazi yang melakukan eksperimen untuk kepentingannya. Lama setelahnya, Dr. Charles Xavier (McAvoy) dan Erik Lensherr (Fassbender), bersama rekan-rekan mutan lainnya harus bekerjasama melawan Sebastian Shaw (Bacon) beserta komplotannya yang ternyata adalah Schmidt yang ingin melenyapkan umat manusia.

Inti dan kunci kisah filmnya memang pada karakter Erik Lensherr dan Charles Xavier, bagaimana mereka bertemu, bagaimana akhirnya mereka berseteru, serta lahirnya X-Men. Film ini sendiri merupakan prekuel dari tiga seri X-Men sebelumnya dan tentu kelemahan film ini adalah bagi penonton yang belum menonton film-film sebelumnya. Baik karakter-karakternya dan hubungan kausalitas cerita saling bertautan. Kisah film ini sendiri terbagi menjadi beberapa sub plot yang lumayan rumit dan kompleks melibatkan puluhan karakter baik mutan dan manusia. Semuanya terangkai dengan baik nyaris tanpa cacat untuk sebuah prekuel. Hanya siapa sangka ternyata Raven (Mistique) dulunya begitu dekat dengan Xavier (bak kakak adik) tanpa memperlihatkan satu saja kedekatan emosional pada tiga seri X-Men sebelumnya.

Kasting pemain adalah kunci keberhasilan utama film ini. Pilihan James McAvoy dan aktor non bintang asal Jerman, Michael Fassbender sebagai “Prof. X” dan “Magneto” adalah pilihan sempurna melihat permainan akting serta chemistry diantara keduanya. McAvoy menampilkan sosok Xavier muda yang cerdas, “playboy”, dan berkesan selalu tak serius dengan sense of humor-nya namun memiliki jiwa memimpin yang tinggi. Lalu Fassbender menampilkan sosok Erik muda yang penuh dendam dan amarah, keras hati, namun sebenarnya memiliki hati yang lembut. Permainan akting keduanya berhasil menjawab kedekatan mereka pada tiga seri X-Men sebelumnya yang dekat layaknya saudara kandung namun berbeda prinsip.

Seperti film-film superhero kebanyakan adegan aksi seru yang penuh efek visual menjadi daya tarik utama filmnya. Sekalipun dengan setting cerita masa lalu (era perang dingin) tetap saja mampu menampilkan sekuen aksi yang memukau dan meyakinkan. Lihat bagaimana Erik mengobrak-abrik sebuah kapal pesiar besar dengan rantai kapal serta tentu sekuen klimaks film. Ilustrasi musik, khususnya musik tema “Erik /Magneto” yang paling dominan, sangat pas mendukung aksi-aksi Erik dan mendukung film secara keseluruhan. Secara umum, X-Men: First Class adalah yang terbaik dibandingkan film-film X-Men lainnya karena mampu memadukan unsur dramatik cerita yang telah ada (seri sebelumnya) dengan efek visual yang memukau. Parapecinta film tinggal menunggu saja film-film X-Men lainnya.
